摘要
目的研究利心冲剂对心衰大鼠基质金属蛋白酶组织抑制因子(TIMP-1)和转化生长因子β(TGF-β)的影响。方法50只Wistar大鼠,随机取10只为正常组,余40只腹腔注射阿霉素(ADR)4mg/kg,每5天1次,共3次,复制大鼠慢性心力衰竭模型。造模成功后分为模型组、阳性药物组、利心冲剂低剂量组、利心冲剂高剂量组。阳性药物组、利心冲剂低剂量组、利心冲剂高剂量组大鼠于造模成功后次日灌胃给药,连续给药21d后,采用Real time PCR检测B型利钠肽(BNP),亲和组织化学技术三步法(SABC法)免疫组织化学检测TIMP-1、TGF-β的表达。结果与模型组比较,利心冲剂低剂量组、高剂量组治疗后胶原沉积明显减少,以高剂量组最为明显;BNP、TGF-β表达明显下降(P<0.01),以高剂量组为显著;TIMP-1的表达明显增加。结论利心冲剂调控TIMP-1、TGF-β信号通路,抑制胶原沉积,改善心功能。
Objective To study the effects of "Lixin Granule" on tissue inhibitor of metalloproteinase-1 (TIMP-1) and transformation growth factor β(TGF-β) in rats of chronic heart failure due to Adriamycin. Methods Fifty rats were randomized into normal group, model group, control grnup, luw-duse " Lixin Granule" group and high-Jose "Lixin Granule" group. Rat model was duplicated by intraabdominal injection of Adrlamycin at 5 mg/kg, once every five days for total three times. Brain natriuretic peptide(BNP) was detected by Real Time PCR, TIMP-1 and TGF-β expressions by SABC immuno-histochemical method. Results Compared with model group, collagen sedimentation was obviously reduced in "Lixin Granule" groups, especially in high-dose "Lixin Granule" group; BNP and TGF-β expressions were evidently reduced(P 〈0.01 ), especially in high-dose "Lixin Granule" group; TIMP-1 expression was increased. Conclusion "Lixin Granule" can regulate TIMP-1 and TGF-β signal pathway and suppress collagen sedimentation to improve the heart functions.
