摘要
目的:建立从麦冬中制备麦冬皂苷D对照品的方法。方法:用萃取、硅胶柱层析、凝胶柱层析、重结晶等方法对麦冬乙醇提取物进行分离纯化,通过MS、IR、1H-NMR、13C-NMR进行结构鉴定。结果:薄层色谱法(TLC)、高效液相色谱法(HPLC-ELSD)检测分析,麦冬皂苷D的纯度为99%以上。结论:本方法简便,产品纯度高,该对照品可作为川麦冬及其制剂质量的指标成分。
Objective:To establish a method for preparing the reference substance of ophiopogonin D from the tuber roots of Ophiopogon japonicus, nethods:Ophiopogonin D was isolated and purified from the ethanol-extract of the tuber roots of Ophiopogon japonicus by solvent extraction, silica gel sephadex LH-20 column chromatography, and crystallization. Its tructure was determined by spectroscopic methods (MS, IR, ^1H- and ^13C-NMR). The purity of the reference substance was identified with TLC and HPLC-ELSD. Results: TLC and HPLC analysis showed that the purity of ophiopogonin D was not less than 99%. Conclusion: The method developed is simple and effective for producing product with high purity. The product obtained can be used as a reference substance in the quality control and the research of herbal medicine.
