摘要
目的观察2型糖尿病大鼠肺组织细胞凋亡率、Caspase-3、Caspase-9的表达变化及罗格列酮的干预作用。方法8周龄SD大鼠高热量饮食1月后,腹腔注射小剂量链脲佐菌素(STZ30mg/kg),成模后分别于12、24周将动物处死,采用光镜、原位末端标记法检测肺组织细胞凋亡及用免疫组织化学(immunohistochemistry)方法检测各组肺组织Caspase-3、Caspase-9表达水平。结果糖尿病大鼠肺组织凋亡细胞数明显增多,Caspase-3、Caspase-9蛋白表达显著上调,但无统计学意义。罗格列酮干预组凋亡率降低(P<0.01),升高的Caspase-3、Caspase-9蛋白表达下调,但与糖尿病未治疗组均无统计学差异(P>0.05)。结论肺组织细胞凋亡在糖尿病大鼠肺损害病程中起一定作用,罗格列酮可抑制肺组织细胞凋亡,可能对糖尿病肺有保护作用。
Objective To observe the apoptotic rate of lung tissue, the change about the expression of Caspase-3, Caspase-9 in type 2 diabetic rats, and the interfering function of rosiglitazone. Methods Feed 8-week-old SD rats with high-calorie diet for one month, then gave them low-dose streptozotocin (STZ 30mg/kg) injection by intraperitoneal route to set -up the T2DM model. Killed the rats at 12, 24 week, respectively. The TdT-mediated nick end labeling and the immunohistochemistry were used to detect the apoptosis of lung cells and the expression of Caspase-3, Caspase-9.Results The number of apoptotic cells in lung tissue was increased, and the expression of Caspase-3, Caspase-9 protein was upregulated, which didn't show significant statistic difference, however. The apoptotic rate of lung cell decreased in rosiglitazone group(P〈0.01), and the up-regulated expression of Caspase-3, Caspase-9 protein was depressed in rosiglitazone group which didn't show significant difference (P〉0.05).Conclusions Apoptosis of lung tissue may play a role for the lung- injury process of diabetic rats. Rosiglitazone may depress the apoptosis of lung tissue, so it may potentially protect the diabetic lung.
