摘要
采用聚丙烯酰胺凝胶垂直平板电泳技术,对选自中国芝麻资源7个生态区的36个地方品种进行了酯酶、过氧化物酶、苹果酸脱氢酶、6—磷酸葡萄糖脱氢酶、淀粉酶5种同工酶分析,仅酯酶酶谱表现出品种间和品种内的多样性差异。对芝麻核心种质和非核心种质中各9个品种的1080个植株酯酶同工酶分析,结果表明在迁移率Rf0.45~0.67间检测到8种酶谱类型。通过对核心种质与非核心种质的酯酶酶谱类型和酶带分布频率、品种多样性指数比较及聚类分析,表明核心种质良好地代表了非核心种质。
The isozyme technology of polyacrylamide gel vertical slab electrophoresis was used to analyze esterase, peroxidase, malate dehydrogenase, 6—phosphoglucoates dehydrogenase and amylase of 36 landraces selected from seven ecological regions of Chinese sesame germplasm resources. The research indicated that only esterase zymograms presented the diversity differences among accessions and within an accession. Total 1080 individuals from nine landraces in core collection and nine in reserve collection originated from three sesame ecological regions of north, central and south China were tested by esterase. The results showed that eight zymogram types were detected out in mobility (Rf) between 0.46 and 0.67. Throuth the comparison of zymogram type frequency, band distribution, varietal diversity index and cluster analysis, the core collection representated the reserve collection well.
