氧化应激下大鼠肝星状细胞增殖情况及硫化氢含量的变化 被引量:1

THE CHANGE OF THE PROLIFERATION OF RAT HSC AND THE CONCENTRATION OF H_2S UNDER OXIDATIVE STRESS

摘要 目的了解氧化应激(Oxidative stress)下肝星状细胞(Hepaticstellate cell,HSC)的增殖情况以及细胞中硫化氢(Hydrogen sulphide,H2S)含量的变化,初步探讨H2S在肝纤维化(Hepatic fibrosis)中的作用。方法利用铁超载体外培养细胞的氧化应激模型,给予不同浓度的外源性H2S供体NaHS和KATP通道阻断剂格列本脲(glibenclamide,GLBN),用MTT检测各组细胞的生长曲线;用敏感硫电极检测各组细胞上清液中H2S含量的变化。结果给予FeNTA后HSC增殖,其OD值随着NaHS浓度的增加和时间的延长而下调;给予GLBN后细胞增殖情况与前者相反;细胞增殖下调时细胞上清液和裂解液中的H2S含量升高,细胞增殖上调时H2S含量降低。结论H2S在氧化应激下能抑制HSC增殖,具有细胞保护作用。 Objective To clarify the change of proliferation and H2S-concentration of HSC under oxidative stress,in order to explore the effect of H2S on hepatic fibrosis.Methods At first a oxidative stress model was established by incubating rat hepatic stellate cells with ferricnitri-lotriacetic acid(Fe-NTa),to supply sodium hydrosulfide(NaHS,H2S donor) and GLBN(a inhibitor of KATP channel through which H2S reacts).The HSC growth curve were detected by MTT,the content of H2S in culture supernatant and cell lysis solution were detected by sensitive sulf-electrode assy.Results The proliferation of hepatic stellate cells with FeNTA interference were increased.The OD was decreased,this trends was accordant with that to add the content of NaHS and to lengthen action time,but it was completely different after GLBN given.It was increased that the content of H2S in culture supernatant and cell lysis solution when the proliferation of HSC was increased,vice versa.Conclusion H2S may inhibit the proliferation of HSC and possess cell protecting effects under oxidative stress.
