摘要
本实验用不连续、非变性聚丙烯酰胺凝胶电泳系统，以损伤坐骨神经远侧端提取的具有神经诱向生长作用的、PI为5．2的18kD蛋白作为抗原，免疫BALB／C小鼠，通过杂交瘤技术，Elisa法筛选，获得二株（Ⅲ8G、Ⅳ3C）稳定分泌单克隆抗体的杂交瘤细胞株。免疫印迹结果表明单克隆抗体能特异性识别分子量为18kD蛋白。免疫组化法显示：在实验组坐骨神经远侧端为阳性反应，对照组坐骨神经的神经膜为阴性。
The study of neuro-chemotropism was an important aspect of neurosciences. The monoclonal antibodies (two lines,Ⅲ8G, Ⅳ3C) were developed through immunizing the BALB/C mouse with 18 kD protein by assay of Elisa. With western blotting, 18 kD McAb can recognize the band of 18 kD protein. Using immunohistochemical method, positive products were seen in the distal segment of the sciatic nerve of experimental mice , but were not in the normal sciatic nerve.
