摘要
采用梯度洗脱HPLC定量,研究乙醇浓度、物料比、提取时间、提取温度和重复提取次数对豆粕提取物中大豆异黄酮6种主要成分得率的影响,为不同大豆异黄酮糖苷或苷元的提取提供理论依据。乙醇浓度对其豆粕提取物中的大豆甙、黄豆黄甙、染料木甙、大豆甙元、黄豆黄素和染料木素的得率影响不显著(P>0.05),乙醇浓度达到60%以上时,6种异黄酮均接近最大值,但70%为利于精制的最低浓度;物料比对3种苷元得率的影响不显著(P>0.05),当物料比大于1∶15(W∶V)时,大豆甙、黄豆黄甙和染料木甙的得率得到显著提高(P<0.05);同提取时间和重复次数相比,提取温度对得率的影响最大,其次是提取时间。这些条件的最佳搭配为70%乙醇,物料比1∶15(W∶V),温度90℃,时间3h,提取1次。
The effects, such as ethanol concentration, ratio of substrate to solvent (RSS), extraction time, temperature and repetition on the extraction of 6 main soybean isoflavones were studied by HPLC analysis. There was no significant influence for ethanol concentration on the yield of daidzin, glyeintin, genistin, daidzein, glycitein and gentistein (p〉0.05). When the ethanol concentration was above 60%, the yields of all the 6 isoflavones were approaching the maximum. Refine was favoring under 70% ethanol. There was no significant effect for RSS on the extraction of 3 agtyeons (p〉0.05), while 1 : 15(W : V) was the best for the extraction of 3 glucosides. Among extraction time, temperature and repetition, temperature was the most important factor, then time. The optimized condition was 70% ethanol, RSS 1: 15,(W : V) 90 ℃, 3 h, repeat 1.
