摘要
目的:建立测定家犬血浆中藁本内酯的HPLC含量测定方法,并用于药动学研究。方法:测定家犬外用与静脉注射给药后藁本内酯的血药浓度,采用药动学程序3P87进行统计处理,计算药动学参数。结果:贴片中藁本内酯的体内过程符合二室模型,其药物动力学参数为Tmax=3.6891h,Cmax=2.2366μg.mL-1,AUC=57.2052μg.h.mL-1。结论:该药具有维持稳定血药浓度时间长、药效稳定的优点。
To establish a HPLC method for the determination of ligustilide, which was applied to the pharmacokinetie study, the concentration of ligustilide in dogs was determined after transdermal gel adminitration and intravenous administration. The pharmacokinetic parameters of ligustilide were analyzed by 3P87. The results obtained indicated that the patch was a two-compartment model, and its druggery dynamics parameters were ： Tmax = 3. 6891 h, Cmax= 2.2366 μg·ml^-1 , AUC = 57. 2052μg·h·ml^-1. It is concluded that this drug has the advantage of long time stable blood-drug concentration and steady effect.
