摘要
采用微生物法,对16名男性健康志愿者单剂口服国产阿奇霉素胶囊后的药代动力学进行了研究。口服后不同时间收集血、尿标本。检测菌为藤黄八叠球菌28001。测定结果用3P87软件经微机处理。结果:口服本品后药时曲线符合二室模型,主要药动学参数,峰时间为(196±078)h,峰浓度为(102±040)mg/L,药时曲线下面积为(1626±319)mg·h·L-1,消除半衰期为(4741±533)h,192h尿累积排出率为(1311±229)%。
The pharmacokinetics of domestic azithromycin was studied in 16 male healthy volunteers who received azithromycin capsules of 1 000 mg orally.Serum and urine samples were collected at various time after oral administration.The serum and urine concentrations of azithromycin were determined by microbiologic method.The used microorganism was Sarcina Lutea 28001.The data of serum level time were processed with 3P87 software.The results showed that the data of domestic capsules conformed to two compartment open model.The main pharmacokinetic parameters,such as t max , c max , AUC and t 1/2β ,were (1.96 ±0.78) h,(1.02±0.40) mg/L,(16.26±3.19) mg·h·L -1 ,(47.41±5.33) h,respectively.The accumulatively excretive rate of azithromycin from urine was (13.11±2.29)% within 192 h.The results indicate that the pharmacokinetics of domestic azithromycin is similar to that of imported azithromycin.
