摘要
聚谷氨酸是一种生物可降解的大分子物质,广泛应用于医药、化妆品等行业。本研究从高温处理过的糖蒜、果仁黄瓜、甜酱黄瓜、蜜汁乳瓜、黄豆酱和海会寺腐乳五种传统酱类食品中分离到高产聚谷氨酸(γ-PGA)菌株,通过对菌落表观形态特征以及生理生化性能的观察分析,初步鉴定为革兰氏阳性枯草芽抱杆菌。该细菌最适培养条件为蛋白胨2%,葡萄糖3%,谷氨酸钠3%,pH7.0,温度37℃,摇床培养250r/min。发酵产品经初步纯化和红外光谱鉴定,产物图谱与标准品图谱基本一致。
γ-Polyglutamic acid (y-PGA) is a kind of biodegradable and giant molecule material which is widely used in medicine and cosmetic. A high γ-PGA-production strain was screened from five kind of china traditional food by high temperature treatment. Through the observation and analysis of the morphology, physiological and biochemical characteristics, the strain was elementarily indentified as Bacillus subblis,gram-positive. The optimum temperature for this bacterium is 35-38 ℃ and the optimum fermentation conditions were: peptone 2%, glucose 3%, glutamic 3%, pH 7.0, temperature 37 ℃, and shaking speed 250r/min. The product was further confirmed by Fuorier transrfrom infrared spectroscopy as γ-PGA.
