摘要 目的考察阳离子交换树脂法中不同洗脱剂对苦参总碱的纯化效果,优选最佳纯化工艺。方法以苦参碱、氧化苦参碱和总生物碱的洗脱率作为考察指标,分别考察洗脱剂(氨水、氨性乙醇、盐酸、氯化钠溶液)对苦参总碱纯化精制的影响,并优选最佳工艺参数。结果四种洗脱剂对苦参总碱的纯化效果差异较大:3mol?L-1氯化钠溶液效果最优,盐酸溶液次之,氨性乙醇溶液居中,氨水溶液最差。结论以氯化钠溶液作为洗脱剂纯化苦参总碱时效率较高,简便、快捷,可用于苦参总碱的纯化。 Objective To select the optimum purifying process of total alkaloids in Sophora flavescens by different eluents on cation exchange resin. Methods Different eluents such as ammonia water, ammonia water with ethanol, sodium chloride and hydrochloric acid were researched, with elution rate of matrine, oxy-matrine and total alkaloids as the indexes , to choose the optimal process parameter. Results The results indicate that the elutropic ability for total alkaloids from Sophoraflavescens is in the following order: 3 mol·min^-1 sodium chloride 〉 hydrochloric acid 〉 ammonia water with ethanol 〉 ammonia water. Conclusion The 3 mol·L^-1 sodium chloride as eluent has high efficiency .It can be used for purifying total alkaloids from Sophoraflavescens by cation exchange ion resin.
