Ghrelin对急性胰腺炎大鼠GH和IGF-1及HGF轴的影响

摘要 目的探讨Ghrelin是否通过生长激素(GH)、类胰岛素样生长因子-1(IGF-1)、肝细胞生长因子(HGF)内分泌轴对急性胰腺炎大鼠起到治疗作用。方法将Wistar大鼠54只随机分为假手术组、急性胰腺炎组、Ghrelin治疗组。分别于术后6、12、24h处死,测定血清GH、IGF-1、HGF水平,检测小肠组织含水量、转运系数及病理。结果模型组与Ghrelin治疗组在小肠含水量、IGF-1水平及小肠病理评分上差异无统计学意义(P>0.05)。Ghrelin治疗组转运系数均高于模型组(P<0.05)。24h3组GH水平比较差异无统计学意义(P>0.05)。在12h及24hHGF水平Ghrelin治疗组与模型组比较差异无统计学意义(P>0.05)。结论Ghrelin可能并不是通过GH-IGF-1-HGF内分泌轴起到治疗胰腺炎作用的。 Objective To investigate the therapeutic effect of ghrelin on acute pancreatitis in rats, whether that is through GH-I-GF-1-HGF axis or not. Methods 54 Wistar rats were randomly divided into 3 groups, sham operation group, AP model group, Ghrelin treatment group. The 6h, 12h,24h after cessation of sodium taurocholate injections the rats were killed, and the levels of serum GH, IGF-I, HGF were detected, and the small intestines were taken out to observe the pathological changes, transit index and water content of small intestines. Results There was no significant difference in the pathological changes between ghrelin treatment group and AP Model group, neither was the water content of small intestine and IGF-1 ( P 〉 0.05 ). The transit index in ghrelin treatment group was higher than that of SAP model group( P 〈 0.05 ). There was no significant difference in the levels of GH among the 24h sub-groups ( P 〉 0.05 ). There was no significant differences in the levels of HGF between ghrelin treatment group and sever acute pancreatitis group on 12h and 24h( P 〉 0.05 ). Conclusion The therapeutic effect of ghrelin is independent,which is not related with GH-IGF-1-HGF axis.
