摘要
目的研究小檗胺与氟他胺联合应用对雄激素依赖性前列腺癌细胞株LNCaP的抑制作用。方法以不同浓度的小檗胺作用于LNCaP细胞株24、48、72h,并以氟他胺及小檗胺联合氟他胺作为对照,采用噻唑盐(MTT)法检测小檗胺和氟他胺对LNCaP细胞的抑制作用,流式细胞术检测细胞周期分布,并在透射电镜下观察细胞超微结构改变。统计分析应用SPSS 11.0软件包,组间比较行单因素方差分析,细胞周期结果采用Х^2检验。结果MTT结果显示,40mg/L小檗胺作用24、48、72h对LNCaP细胞的抑制率分别为(34.43±1.92)%、(63.72±2.46)%、(83.42±2.02)%,与对照组比较差异有统计学意义(P〈0.01);小檗胺对LNCaP细胞的增殖抑制作用呈时间、浓度依赖关系。小檗胺联合氟他胺对LNCaP的增殖抑制作用具有协同性。透射电镜下见经小檗胺作用后细胞具有典型的凋亡形态学改变;流式细胞仪检测LNCaP细胞有明显的凋亡峰,凋亡强度具有时间、浓度依赖性。结论小檗胺通过影响细胞周期和诱导细胞凋亡等机制抑制前列腺癌细胞株LNCaP的增殖,并具有时间和浓度依赖性;小檗胺联合氟他胺对LNCaP的增殖抑制作用具有协同性。
Objective To study the effect of berbamine in combination with flutamide on human prostatic cancer LNCaP cells in vitro. Methods By utilizing the technique of culture in vitro, androgen-sensitive LNCaP prostate cancer cells were treated with different concentrations of the berbamine for 24, 48, 72 h. In the control groups, the LNCaP cell lines were treated with flutamide and flutamide with berbamine. The changes of cell morphology, apoptosis were observed by light microscope, transmission electron microscope (TEM), MTT and flow cytometry. Results After applying 40 mg/L berbamine on LNCaP cell for 24, 48, 72 h, the inhibition ratio were (34.43±1.92)%, (63. 72±2.46) %, (83. 42±2.02) % respectively(compared with the controls, P〈0.01). The result of MTT showed that berbamine had obvious effects on the inhibition of cell proliferation of LNCaP cells, and the effect showed in a time-and-concentration-dependent manner. The combining use of ber- bamine and flutamide had evident synergistic effect on the inhibition of cell proliferation of LNCaP cell. The cells affected by berbamine had typical morphology changes of apoptosis by TEM. After the cells were treated with berbamine, the apoptotic peak could be detected obviously by flow cytometry,and the apoptotic intensity showed in a dose-time dependent manner. Conclusions Berbamine can inhibit the proliferation of LNCaP prostate cancer cells by affecting cell cycle and inducing apoptosis in a time-and concentration-dependent manner. The combining use of berbamine with flutamide has a synergistic action in inhibiting the proliferation of LNCaP.
