不同切片和染色技术在陶瓷支架体内植入中的组织学研究 被引量:4

摘要 目的探讨石蜡切片与硬组织切片及不同染色方法在体内组织工程研究中的应用。方法将大节段多孔生物陶瓷/聚乳酸复合支架植入犬4个不同的非骨部位,取材后分石蜡切片组和硬组织切片组做HE和M asson染色,在光镜下观察其组织学形态并做比较。结果石蜡切片组色彩鲜艳,可以清楚的观察到成骨细胞、类骨质等成分;硬组织切片组镜下观察未充分体现出其优越性。结论M asson染色是HE染色的有益补充,两者结合更有利于镜下组织形态学观察;硬组织切片工序多、难度大,质量控制尚需完善,在本实验研究中,石蜡切片仍是非常重要和有效的观察手段。 Objective To study the applications of paraffin sections, hard tissue slicing and different methods of staining in the study of tissue engineering in vivo. Methods We implanted large segment of porous bio-ceramic in four different non-bone parts of dogs, drew biopsies from them and divided biopsies into group of paraffin sections and group of hard-tissue slicing. The two groups were stained by HE and Masson. Then the histological patterns were observed under microscope and compared. Results The paraffin sections were brightly stained. And the osteoblasts, osteoid and other ingredients could be recognized clearly under microscope. The expression in hard tissue slicing group was not superior to others. Conclusion Masson staining was a beneficial addition for HE staining. A combina- tion of both was more favorable for tile observation of histological patterns under microscope. The procedure of hard tissue slicing was more and more difficult than the procedures of paraffin sections, and the control of quality was not perfect. In our study, paraffin sections were very important and effective means of observation.
