瘢痕成纤维细胞整合素表达实验研究 被引量:8

摘要 目的:了解在体外培养的瘢痕及正常皮肤组织来源的成纤维细胞整合素表达情况。方法:应用β1及α1~α4整合素单克隆抗体对成纤维细胞进行整合素原位ELISA检测,观察不同培养时期瘢痕成纤维细胞的β1及α1~α4整合素表达水平,辅以正常皮肤成纤维细胞做对照。结果:培养的瘢痕成纤维细胞整合素各亚型表达均明显高于同期培养的正常皮肤成纤维细胞表达水平(P<0.01);细胞经较长时间培养后,整合素的表达均有所下降,但就下降幅度而言,瘢痕成纤维细胞为5.94%~18.26%,平均为9.75%;而正常成纤维细胞为26.19%~46.84%,平均为32.46%;两者有显著性差异(P<0.01)。结论:瘢痕成纤维细胞具有比正常皮肤成纤维细胞更强的整合素表达功能,从而对瘢痕组织增生挛缩的发生。 Objective: To realize expression levels of integrins in hypertrophic scarderived and normal skinderived fibroblasts. Methods: Using anti-β_1, α_1, α_2, α_3 and α_4 integrin McAbs, the expression of integrins was detected in hypertrophic scarderived and normal skinderived fibroblasts of passage 5 and 15 by ELISA technique. Results: The hypertrophic scarderived fibroblasts possessed higher expression level of integrin subunits than normal skinderived fibroblasts. After cells were cultured for long time, the decreased range of integrin expression in hypertrophic scar-derived fibrolasts was 5.94%~18.26%, and that in normal skinderived fibroblasts was 26.19%~46.84%. There was a statistical difference between them (P<0.01). Conclusion: Overexpression of integrins in the hypertrophic scar fibroblasts may play an important role in the hypertrophic scar forming and be contemporaneous with tissue contracture.
