摘要
采用胚胎肢芽细胞微团培养方法,观察无机氟对SD大鼠和昆明种小鼠胚胎肢芽细胞增殖和分化的影响。结果显示培养液中一定浓度的氟对两种动物胚胎肢芽细胞增殖和分化有明显抑制作用,且呈剂量-反应关系。拟合细胞分化和增殖抑制曲线,计算得大、小鼠半数抑制分化浓度(ID50)分别为6.8μg/ml、7.3μg/ml;半数抑制增殖浓度(IP50)为44.1μg/ml、63.6μg/ml。IP50/ID50分别为6.4和8.7。表明无机氟是肢芽细胞分化的特异性抑制剂。
The effect of fluoride on differentiation and proliferation of rat and mouse embryo limb bud cell were studied with micromass cultures in vitro. Embryo limb bud cells of rat (13day) and mouse (12day)were subjected to culture for 5 days. The results showed that fluoride could inhibit differentiation of cells without affecting cells proliferation. The concentrations of 50% inhibition of cell differentiation (ID50) were 68μg/ml(rat) and 73μg/ml(mouse). The concentrations of 50% inhibitions of cell proliferation (IP50) were 441μg/ml (rat) and 636μg/ml (mouse). The IP50/ID50 values 64(rat) and 87 (mouse) were both greater than 5. According to the assessment criteria of Flint and Cheng Wanrong, the fluoride may be an embryo limb bud cells specific inhibitor. It could have potent teratogenicity.
