摘要
目的观察隐神经部分结扎(saphenous partial ligation,SPL)大鼠背根神经节P2X3受体表达的改变。方法麻醉下,制备大鼠SPL模型;测定SPL大鼠机械异常性疼痛和热痛敏的变化;应用免疫组化技术观察SPL大鼠损伤相应节段(L3、L4)的背根神经节P2X3受体表达的变化。结果SPL大鼠可产生热痛敏和机械异常性疼痛;SPL术后第7、14d,损伤同侧的L3、L4节段背根节P2X3免疫阳性神经元数量明显增加。结论SPL是一种适合于病理性神经痛机制研究的模型,SPL可导致损伤同侧相应节段的背根节神经元P2X3受体表达增加。
Objective To observe the alteration of P2X3 receptor expression in dorsal root ganglion in rats of saphenous partial ligation (SPL). Methods Under anesthesia, the model of SPL was performed in SD rats. Mechanical allodynia and thermal hyperalgnsia were evaluated in rats of SPL. P2X3 receptor expression in L3 and L4 DRG following SPL were observed by using a polyclonal antibody to label the P2X3 receptor. Results Rats of SPL developed mechanical allodyaia and thermal hyperalgesia compared with sham animals. A significant increase in P2X3 immunoreactivity was observed in the ipsilateral(injured) L3 and L4 DRG on 7, 14 d after SPL.Conclusion SPL is an adequate model that will provide a new tool for clarifying mechanisms of neuropathic pain. SPL significantly increase P2X3 receptor expression in dorsal root ganglion.
