橙皮苷抑制长波紫外线诱导的人角质形成细胞氧化损伤和凋亡 被引量:2

摘要 目的:探讨橙皮苷是否能对长波紫外线(UVA)诱导的人角质形成细胞(HaCaT细胞)氧化损伤和凋亡起保护作用以及其可能的机制。方法:HaCaT细胞分成空白组、橙皮苷组、UVA照射组以及UVA照射+橙皮苷组。四甲基偶氮唑蓝(MTT)法检测细胞活性,筛选最佳药物浓度;比色法检测细胞上清液中的超氧化物歧化酶(SOD)和总抗氧化能力(T-AOC)活性以及丙二醛(MDA)含量的变化;流式细胞仪检测细胞膜电位变化和不同组细胞凋亡率。结果:UVA照射组的SOD和T-AOC明显下降,MDA上升,线粒体膜电位下降,凋亡率上升(P<0.05)。橙皮苷组细胞照光后其活性以及上清液中的SOD、T-AOC活性相对于未加药照光组明显增加,MAD下降,线粒体膜电位上升,细胞凋亡率也明显下降(P<0.05)。结论:橙皮苷可以减少UVA诱导的HaCaT细胞氧化损伤和细胞凋亡,其机制可能与增强抗氧化能力和减少氧自由基有关。 Objective: To investigate whether hesperidin(HPD) may protect human keratinocytes against oxidative damage and apoptosis induced by UVA irradiation and the possible mechanisms. Methods: The cultured HaCaT cells were divided into four groups: the blank control group, HPD-treated group, UVA-irradiation group, and UVA-irradiation incorporated with HPD-treated group. HaCaT cells were treated by 10 J/cm2 UVA irradiation. The viability of the cells was detected using a tetrazolium-based colorimetric assay. The activity of superoxide dismutase (SOD), total antioxidative capacity (T-AOC), and the level of maleic dialdehyde (MDA) were detected with colorimetric methods. The changes of cell membrane potential and the apoptosis rates of the cells were detected by flow cytometry. Results: After UVA irradiation, the viability, the activity of SOD, the T-AOC and the cell membrane potential of HaCaT cells decreased, while the content of MDA in HaCaT cells and the apoptosis of HaCaT cells increased (P 〈 0.05). However, compared with those treated by UVA irradiation alone, the viabil- ity, the activity of SOD, the T-AOC and the cell membrane potential of HaCaT cells increased in the group of UVA-irradiation incorporated with HPD-treating, accompanied by a decrease of the MDA level and the apoptosis rate of HaCaT cells (P 〈 0.05). Conclusion: HPD can relieve the oxidative damage and the apoptosis of HaCaT cells induced by UVA irradiation, which may be related to the strengthening of antioxidation and the decrease of free radical of oxygen.
