摘要
目的探讨VERGE蛋白在小鼠大脑中动脉梗死模型(MCAO)中蛋白表达的变化及其机制。方法C57BL/6小鼠30只,随机分为2组:30 min MCAO模型组、90 min MCAO模型组,制备MCAO模型,应用激光多普勒血流仪进行术中脑血流监测,应用Western印迹法观察各组梗死侧及非梗死侧的VERGE、p-JNK2、p-ERK1/2、P-38蛋白表达的变化。结果30 min、90 min MCAO模型组梗死侧VERGE、p-JNK2的蛋白表达明显增高,p-ERK1/2、P-38蛋白表达无变化;两组MCAO模型非梗死侧的VERGE、p-JNK2、p-ERK1/2、P-38蛋白表达均无变化。结论VERGE蛋白表达在小鼠MCAO的早期明显升高可能与JNK2通路有关而与ERK、P38通路无关。
Objective To study the protein expression and its mechanism of VERGE in MCAO mice models.Methods The mice were divided into 30 and 90 min MCAO groups.All mice were made into MCAO models.The protein expressions of VERGE,p-JNK2,p-ERK1/2,P-38 were tested by Western blot.Results The protein expressions of VERGE,p-JNK2 of both groups in ischemia hemisphere were increased,while the protein expressions of p-ERK1/2,P-38 had no change.There were no protein expression changes of VERGE,p-JNK2,p-ERK1/2,P-38 in the nonischemic hemisphere of both groups.Conclusions The protein expression of VERGE is increased in the transient MCAO models,the mechanisms of which may be related to JNK2.
