放射性碘-125辐射对液态AFP基因结构的影响

Influence of radiation of radioactive iodine-125 on DNA structure of liquid AFP

摘要 目的:探讨放射性碘-125(125I)辐射强度与DNA损伤的关系.方法:微量法激光拉曼光谱检测被不同浓度125I发射的俄歇电子辐射损伤液态甲胎蛋白基因(AFPDNA),分析DNA构象及碱基的拉曼光谱变化特点及特征峰相对强度变化规律.结果:125I发射的俄歇电子辐照AFP基因后,可见核糖、磷酸键及主链C-C构象的拉曼振动光谱峰偏移及峰值降低,并见嘌呤及嘧啶环的功能基团光谱峰降低.随125I浓度增加,逐渐出现DNA片段的特征拉曼光谱峰,并见核糖体暴露及碱基脱落堆积的新拉曼光谱峰.结论:125I辐照AFPDNA可引起DNA构象及功能基团损伤,随辐射强度增加逐渐出现碱基脱落及DNA结构破坏. AIM:To study the relation of the radiated intensity and the DNA damage induced by Auger electron. METHODS: The Alpha-fetoprotein DNA solution radiated by different doses of Auger electron emitting from the radioactive iodine-125 was examined using the Laser Raman spectroscopy. RESULTS: The differentiation of characteristic Raman vibration band of DNA was extensive and its peak intensity reduced after the DNA was radiated by Auger electron in 24 h. The Raman peak of the functional groups of pyridine and pttrine bases, the ribosbme, the phosphate bond and the conformation of DNA backbone became weak. However, the characteristic Raman peak of the DNA segments gradually emerged while the radioactive dose of iodine-125 increased. The pyridine and purine bases were dissociated and stacked and the peak intensity of the Raman spectra increased while the radiation on DNA became stronger. CONCLUSION: Auger electron emitting from iodine-125 may damage the conformation of DNA backbone and DNA structure. The damage extent of DNA is related with the radiated intensity.
