Separation of Protein With Multiple Injection on High-Performance Ion -Exchange Chromatography

摘要 研究了离子交换色谱上的累加进样法,并用选择性累加进样法在SCX-6柱子上纯化了尿激酶粗品,结果较好。 An abrupt change of the eluting curve of proteins in the high-performance ion-exchange chromatography (HPIEC) was reported. When the composition of mobile phase is far away from the position of the abrupt change, the multi-injection of protein before and after starting a gradient elution for one chromatographic separation in the HPIEC cap be performed. Thus the variation of protein capacity factor can not he found significantly. That is to say, the two operation processes, concentration and separation, can be finished on one chromatographic procedure. This method can be used to overcome the difficulty that the sample solution can not be detected and to enhance the analytical sensitivity. We also use the multiple injection method for the purification of crude urokinase. The impure pro- tein goes through foe column with the solution A after each injection. Therefore the effeCtive column ca- pacity is relatively raised.
