摘要
目的:体外培养大鼠胚胎脊髓神经干细胞,观察其生长、增殖和分化特点。方法:利用倒置显微镜、MTT法、免疫细胞化学等方法检测神经干细胞的增殖、分化情况。结果:分离的脊髓神经干细胞可形成大量悬浮的神经球,并可进行体外扩增传代,神经球内的细胞均呈nestin阳性。在添加10%胎牛血清7d后,神经球细胞可分化为有神经元、星形胶质细胞特异性抗原表达的细胞。结论:体外培养大鼠胚胎脊髓可得到大量神经干细胞,并能分化为神经元和星形胶质细胞。
Objective: To isolate and culture neural stem cells from rat embroyonic spinal cord and observe the characteristics during growth, generation and differentiation. Methods: We detected the generation and differentiation of neural stem ceils by light microscope, MTT test and the methods of immunocytochemistry. Results: Cells separated from spinal cord of embryonic rats could proliferate and passage in vitro. Nestin-positive cells were detected in neural sphere. The separated cells cultured in serum with 10% FBS for 7 days might naturally differentiate into cells which strongly expressed neurons or astroeytes antigen at some rate. Conclusion: The above results suggest that there are neural stem cells from rat embroyonie spinal cord, which may differentiate neurons and astrocytes.
