摘要
对除草剂靶标对羟基苯基丙酮酸双氧化酶(HPPD)的研究进展情况作了简要的概述,总结了植物体内HPPD的作用机制和HPPD抑制剂的作用机制,重点以NTBC为例综述了NTBC与对羟基苯基丙酮酸双氧化酶的作用研究来阐明抑制剂对HPPD酶的抑制机理.研究表明抑制剂NTBC与靶标HPPD的强亲和力主要通过两个氧原子与Fe″的二齿配位和蛋白质口袋中芳环间的π-堆积力作用,其中芳环间的π-堆积力起主要作用.
The progress of research on the herbicide target of p-hydroxyphenylpyruvate dioxygenase was reviewed, and summarized the action mechanism of plant HPPD and action mechanism of HPPD inhibitor, mainly summarized the interaction of NTBC to HPPD to illustrate the mechanism of inhibitor to HPPD with NTBC as a main example. The researches indentified two binding interaction of NTBC to HPPD that might relate to its high affinity: bidentate coordination to Fe^Ⅱ and π-stacking between the aromatic ring of NTBC and two phenylalanine residues in the protein pocket, the π-stacking interactions in the protein pocket appeared to be responsible for the high affinity.
