摘要
目的探讨芦丁对大鼠α-葡萄糖苷酶活性的影响。方法体内试验:将30只SD大鼠随机分为对照组,芦丁组和阳性对照阿卡波糖组,一次性灌胃给药测定其餐后60min血糖值的变化。体外实验:将芦丁和阿卡波糖加入到大鼠小肠提取的酶液中,以麦芽糖为底物,用氧化酶法测定葡萄糖的生成量,确定芦丁对α-葡萄糖苷酶的影响。结果芦丁显著降低大鼠餐后60min血糖水平,与对照组比较,差异具有统计学意义(P<0.01),体外实验显示,随着芦丁浓度的增加,吸光度值逐渐减小,其α-葡萄糖苷酶的抑制率逐渐增加。结论芦丁体内外对α-葡萄糖苷酶活性有较强的抑制作用。
Objective To investigate the effect of rutin on activity of α-glucosidase in rats.Methods 30 SD rats were randomly divided into three groups as follows,normal control,rutin and positive acarbose groups.Postprandial blood glucose changes in 60 min were detected after intragastric administration for once.Acarbose and rutin were respectively added to the extracted enzyme solution in vitro,followed by the measurement of glucose value in the substrate of maltose to determine the absorbance of α-rutin glycosidase. Results Rutin had a significant effect on the reduction in blood glucose levels in rats （ P〈0. 01 ） compared with that of the normal group. Rutinin reduced absorbance value, the difference had a statistical significance in vitro. Experiments showed that with the increase of turin concentration, ahsorbance values gradually decreased, and its inhibition rate of α- glucosidase gradually increased, Conclusion Rutin can inhibit the activity of α- glucosidase in vitro and in vivo.
