摘要
目的:建立高效毛细管电泳法测定盐酸赛庚啶片中盐酸赛庚啶含量的方法。方法:采用熔融石英毛细管柱,45cm×75μm(有效长度37cm);运行缓冲液50mmol/L醋酸盐缓冲液(pH4.75);压力进样3.448kPa×5s;工作电压25kV;柱温25℃;检测波长286nm。结果:盐酸赛庚啶浓度在0.02~0.20g/L范围内,该方法线性关系良好,Y^=10.0507X-0.0069(r=0.9995),盐酸赛庚啶的最低检测限为0.002g/L,最低定量限为0.007g/L。精密度、回收率均符合标准。3批样品的相对含量分别为98.75%,99.26%,99.10%。结论:高效毛细管电泳法简便、准确、快速、重复性好,可用于盐酸赛庚啶片中盐酸赛庚啶含量的测定。
Aim: To establish a method for the determination of cyproheptadine hydrochloride by high performance capillary eleetrophoresis (HPCE). Methods: The determination was performed on the following conditions: fused-silica capillary of 46 cm x 75 Ixm ( 37 cm of effective length) ; running buffer : 50 mmol· L^-1acetate buffer ( pH 4.75 ) ; sample injection method :3. 448 kPa x 5 s; running voltage : 25 kV ; column temperature : 25℃ ; detect wavelength: 286 nm. Results : The linear concentration range was from 0.02 to 0.20 g/L of eyproheptadine hydrochloride, the linear function was = 10. 050 7X - 0. 006 9 ( r = 0. 9995 ). The lower detection limit was 0. 002 g/L, and the lower quantity limit was 0. 007 g/L. The RSD of degree of precision and recovery measured up to standard. The relative content of three batches was 98.75% ,99.26% ,99.10% , respectively. Conclusion:HPCE method is simple, accurate, rapid and reproducible. It can be used to detect the content of cyproheptadine hydrochloride tablets.
