食管鳞状细胞癌组织中TPX2的表达 被引量:1

Expression of TPX2 in esophageal squamous carcinoma tissue

摘要 目的:探讨食管鳞状细胞癌组织中TPX2的表达和意义。方法:采用免疫组织化学SP方法检测TPX2蛋白在62例食管鳞癌、31例癌旁不典型增生组织及62例正常食管黏膜组织中的表达。运用RT-PCR分析22例食管鳞癌组织、18例对应的癌旁不典型增生和正常食管黏膜组织中TPX2mRNA的表达水平。结果:正常食管黏膜、癌旁不典型增生及食管癌组织中TPX2蛋白阳性率分别为4.8%(13/62)、51.6%(16/31)和85.5%(53/62),3组间比较差异有统计学意义(χ2=20.037,P<0.01)。癌组织、癌前病变组织中TPX2mRNA阳性率分别为73.3%(16/22)、60.0%(12/18),正常食管黏膜组织中TPX2mRNA几乎不表达,3种组织间比较,差异有统计学意义(P<0.05)。食管鳞癌组织中TPX2蛋白及mRNA的表达与癌组织浸润深度及淋巴结转移有关(P均<0.05)。结论:TPX2高表达可能是食管鳞癌淋巴结转移的危险因素;TPX2有望成为食管鳞癌早期诊断及判断预后的指标。 Aim:To study the expression of TPX2 and its significance in esophageal squamous cell carcinoma(ESCC) tissue.Methods:Immunohistochemical staining was used to detect the expression of TPX2 protein in 62 cases of ESCC,31 cases of adjacent atypical hyperplasia epithelium and 62 cases of normal esophageal epithelium tissue.The expression of TPX2 mRNA in 22 cases of ESCC,18 cases of adjacent atypical hyperplasia epithelium and 18 cases of normal esophageal epithelium tissue were detected using RT-PCR.Results:In ESCC,adjacent atypical hyperplasia epithelium and normal esophageal epthelium tissue, the positive rate of TPX2 protein expression was 85.5% (53/62) ,51.6% ( 16/31 ) and 4.8% (13/62) ,respectively, decreased by turn(χ^2 = 20. 037 ,P 〈 0.01 ) ; the positive rate of TPX2 mRNA expression was 73.3% ( 16/22 ) ,60.0% (12/18) and 0,respectively, decreased by turn(P 〈 0.01 ). The expressions of TPX2 protein and mRNA were correlated with lymph node metastasis and infiltration depth of ESCC (P 〈 0.01 ). Oonclusion : High expression of TPX2 may be a risk faetor for lymph node metastasis,and maybe a potential biomarker for early diagnosis and prognosis of ESCC.
