摘要
以草莓品种M14叶盘为外植体,建立了一个有效的、高频率的芽再生系统。通过对基本培养基、叶龄、基因型、暗培养处理以及激素组合等因子的研究,探讨了这些因子对草莓叶盘不定芽诱导的影响。在MS基本培养基附加3.0mg/LBA和0.2mg/L2.4-D的培养基上,M14叶盘不定芽再生频率达94.44%,平均每叶盘再生芽率10.56个。暗培养处理不能提高M14叶盘的芽再生频率。14~30d叶龄的叶盘有较高芽再生频率。M14叶盘再生芽系统可用作草莓的微繁殖系统和基因转化的受体系统。
An efficient and high frequency method of direct shoot regeneration from leaf disc in strawberry(Fragaria×nanassa,M 14 )has been established.The influences of basal medium,age of leaf,genotypes,dark treatment,horemone condition on shoot generation were examined.Regeneration rate of disks reached 94.44% with an average of 10.56 shoots per leaf disc with in 5 weeks when Ms basal medium supplemented with 3.0mg/L BA and 0.2mg/L2.4-D.Dark treatment did not enhance shoot regeneration of M 14 .The age of leaf disks from 14 to 30 days produced shoot at higher frequency than those from less than 14 days or more than 30 days of leaves.This technique will be applied for rapid propagation and genetic manipalation of strawberry.
