环孢素A对大鼠急性脊髓损伤后iNOS表达及细胞凋亡的影响

摘要 目的探讨急性脊髓损伤后应用环孢素A(CsA)对诱导型一氧化氮合酶(iNOS)表达及细胞凋亡的影响。方法108只SD大鼠随机分为对照组、损伤组和CsA治疗组。使用免疫组织化学EnVision法检测急性脊髓损伤大鼠损失节段iNOS表达,原位末端标记法(TUNEL法)标记细胞凋亡并计算凋亡指数。结果损伤组、CsA治疗组及对照组iNOS表达及凋亡指数经单因素方差分析,差异有统计学意义。结论CsA能抑制大鼠脊髓损伤后iNOS的表达及细胞凋亡,减轻脊髓继发性损伤。 Objective To investigate the effect of cyclosporine A on expression of iNOS and apoptosis after spinal cord injury in rats.Methods One hundred and eight SD mice were divided randomly into injury group,CsA treated group and controlgroup.An experimental rat model of acute spinal cord injury was established.The expressions of iNOS in the rat spinal cord was detected by immunohistochemistry staining,and apoptotic cells in the in the tissue sections were tested by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling(TUNEL)method.The expression of iNOS was analyzed quantitatively,and the apoptotic index was evaluated.Results There was statistically significant difference in the expression of iNOS,and apoptotic index among injury group,CsA treated group and control group.Conclusion CsA depress significantly the expression of iNOS and apoptosis after spinal cord injury,and relieve the secondary spinal cord injury.
