高盐饮食和两肾一夹高血压模型对NO及其合酶的影响 被引量:4

Comparison of Indexes for Endothelium Injuries between High-salt and Two Kidneys One Clipped(2k1c)-Induced Hypertension

摘要 目的:NO合成和分泌减少是内皮损伤的主要特征。本实验用高盐饮食和两肾一夹处理SD大鼠,检测NO及其合酶指标,分析不同因素所致高血压对内皮功能的影响。方法:8周龄SD大鼠32只,随机分组:空白对照组(Ⅰ组)8只;两肾一夹组(Ⅱ组)12只;高盐饮食(8%NaCl饲料,Ⅲ组)12只;3周龄SD大鼠(Ⅳ组)10只,处理同Ⅲ组。检测NO、tNOS、i NOS含量及基因CYP2C6在心脏和肾脏的表达。结果:Ⅱ、Ⅲ组NO、tNOS含量均较Ⅰ组降低(P<0.05),而i NOS含量增加(P<0.05),但Ⅱ、Ⅲ组间各指标含量未见差异(P>0.05)。Ⅳ组的相关指标与Ⅲ组均有差异(P<0.05)。结论:盐性和肾性高血压对内皮损伤程度相似;幼龄大鼠对高盐饮食耐受性强;CYP2C6基因与高血压内皮损伤未见明显关系。 Objective: High-salt diet and 2klc were used to establish rat models with hypertension. We detected endothelium injuries-related indexes,NO,tNOS and iNOS, and thus compared endothelium injuries and extents in the two models with a view to realizing how endotheliurn injures play a role in the genesis of hypertension. Methods: 32 SD rats aged 8 weeks were randomly divided into three groups:control(n= 8),2klc(n=12)and 8% NaC1 diet (n= 12). Another 10 SD rats aged 3 weeks were grouped as the fourth, fed with 8% NaC1 diet as we did to the high-salt diet group. NO, tNOS and iNOS were measured, and CYP2C6 gene was detected. Results:Compared to the control group, contents of NO and tNOS decreased(P〈0. 05) while hNOS increased(P〈0. 05) in all groups. Compared to the fourth group, indexes mentioned above were more obvious in the high-salt diet group(P〈0. 05). However, there were no significant differences in these indexes (P〉0. 05) between high-salt diet group and 2klc group. CYP2C6 gene was invisible in kidney in all groups while it was weakly expressed in heart. Cotaelusion: High-salt diet and 2klc cause hypertension and endotheliurn injuries, but such injuries retain almost the same level in adult rats,while endothelium injuries in young rats are not as obvious as those in adult rats. Endothelium injuries are involved in the genesis of hypertension.
