具有自主复制功能的水稻高度重复序列ARS2的结构和功能分析

摘要 对一个水稻珍汕97A不育系核DNA来源的具有自主复制功能的高度重复顺序片段ARS2(全长4270bp)进行了亚克隆构建和测序,获得了它的全顺序.该片段富含AT(69%),有2个ARS核心顺序(A/TTTTAPuTTTA/T),7个类核心顺序,其上下游20~50bp内有TNTPuAA的ARS特征顺序;并分析得出它的2个ARS功能关键区域(分别长400bp和360bp).同时,还首次发现了具有较高多态性的水稻微卫星DNA(AT)n. Subcloning and sequencing have been performed on a highly repetitive fragmentwith autonomously replicating capability from rice Zhenshan 97A nucleic DNA, whose lengthis 4 270 bp. The DNA segments were sequenced. It has high AT ratio (69%), two ARS core sequences(A/TTTTAPuTTTA/T) and 7 similar ones which comes with ARS characteristic sequence TNTPuAA at its upstream and/or downstream. Two key regions of ARS function are delimited in it with length of 400 bp and 360 bp. A microsatellite of rice locus genome(AT)n with high polymorphism was observed for the first time.
