摘要
通过微弹轰击法转化获得的含有bar基因的转基因燕麦(AvenasativaL.)植株(RO)自交产生转基因后代.利用PCR方法检测bar基因在后代中的传递情况.在两个独立转基因系的后代R1(第1代)群体中,有一个检测到了bar基因的稳定传递.在所有检测的R2(第2代)群体中,bar基因的分离符合孟德尔分离比例.其中一个群体为bar基因纯合体.但R1群体中bar基因分离不符合孟德尔分离比例,可能是由于取样误差造成的.分子杂交进一步确认了PCR的结果.PAT酶(bar基因表达产物)活性检测结果表明bar基因在后代植株中能够稳定表达.
Transgenic progenies were obtained by self-pollinating independent transgenic oat (Ayena sativa L.) lines containing har gene driven by rice Actl gene promoter, produced using microprojectile bombardment. Presence of the bar gene was detected in the R1 (the first generation) and R2 (the second generation) progenies of one of the transgenic lines using the Polymerase Chain Reaction, and was confirmed using dot blot hybridization. A Mendelian segregation was found in all the R2 populations examined, but not in the R1 progeny, which was probably caused by sampling error. One of the R2 populations was found to be uniformly bar gene positive, indicating it being derived from a homozygous R1 plant. A stable expression of the bar gene was detected in both the R1 and R2 progenies. No inactivation of bar gene expression was observed in the progeny populations examined.
