摘要
用PCR-RFLP方法扩增了江浙沪籍汉族人群HLAⅡ类基因DQB1基因的第2个外显子,扩增产物经HaeⅢ,Bsp12861,BssHⅡ,BsaHⅠ,HaeⅡ,ApaⅠ,HpaⅡ,RsaⅠ酶切分型,测定了江浙沪籍汉族人HLAⅡ类基因DQB1的16个等位基因的分布频率,DQB1*0601(43%)和*0602(31%)为最常见的等位基因.统计分析表明,在DQB1各等位基因分布上,江浙沪籍汉族与新疆汉族(46例),北方汉族(92例),及用PCR-SSO方法得出的上海汉族差异明显,与藏族、维族、哈族也具有一定差异.
The second exon of HLA-DQB1 was analyzed using PCR-RFLP genotyping method in Han Chinese populations distributed over Jiangsu, Zhejiang, Shanghai areas. The amplified products were digested by HaeⅢ, Bsp12861, BssHⅡ, BsaHⅠ, HaeⅡ, ApaⅠ,HpaⅡ, RsaⅠ to determine the gene frequencies of all alleles at DQB1. Of the all possible 16 alleles in DQB1, DQB1* 0601 and *0602 are the most frequent alleies in Jiangsu, Zhejiang,Shanghai areas. This study suggests variation in the allelic frequency distribution among the different Chinese Han populations and between the Chinese populations and the populations of Tibetan, Uygur, Kazark.
