摘要
目的:探讨金雀异黄素对氧化型低密度脂蛋白(oxidized low density lipoprotein,ox-LDL)诱导的血管平滑肌细胞中活性氧(reactive oxygen species,ROS)生成的影响。方法:以体外培养的人脐静脉平滑肌细胞(human umbili-cal vein smooth muscle cell,hUVSMC)为对象,用50 mg/L的ox-LDL作用细胞2 h,并加入不同浓度金雀异黄素(10,30,90μmol/L)对其进行干预,应用流式细胞技术和荧光分光光度法分别检测细胞内总ROS水平。结果:hUVSMC经ox-LDL刺激2 h后,细胞内ROS水平明显升高(P<0.01);金雀异黄素(10μmol/L)降低ox-LDL诱导的细胞内ROS的增加;金雀异黄素(30,90μmol/L)则进一步提高细胞内ROS水平。结论:金雀异黄素(10μmol/L)可清除细胞内ROS,而金雀异黄素(30,90μmol/L)则增加细胞内ROS的生成。
Objective: To investigate the effect of genistein on reactive oxygen species (ROS) production in vascular smooth muscle cell (VSMC) induced by oxidized low density lipoprotein (α-LDL). Methods: Human umbilical vein smooth muscle cells were pretreated with genistein in different concentrations ( 10, 30, 90 μmol/L) and then treated by 50 mg/L of α-LDL for 2 hours. The levels of ROS were determined by using flowcytometry and fluorospectrophotometry. Results: The level of ROS in VSMC could he enhanced by α-LDL, genistein( 10 μmol/L) might decrease the level of ROS induced by α-LDL, but when the concentration of genistein arrived at 30 μmoL/L and 90 μmol/L respectively, it could further enhance the content of ROS. Conclusion: Genistein( 10 μmol/L) might remove ROS, but genistein(30, 90 μmol/ L) induced the generation of ROS in VSMC.
