纤维连接蛋白及其整合素受体在人胚胎和成熟肾中的表达 被引量:6

摘要 为探讨纤维连接蛋白(FN)及其β1整合素受体在人胎儿和成熟肾中的表达及意义,采用免疫组化的方法观察纤维连接蛋白及其整合素受体(α5β1)的α5亚单位在胎儿和正常成熟肾组织中的分布。结果发现:在胚胎肾组织中,FN及α5在输尿管芽细胞周围有弱阳性表达,在肾小球发育早期,FN及α5在肾单位中表达较弱,其在间质网状分布,当血管插入肾单位后,FN表达明显增加,主要分布于系膜区和毛细血管壁。成熟肾组织中FN及α5在肾小球基底膜(GBM)强阳性表达且以血管极节段明显,间质及小血管壁亦为阳性。结果提示,FN和整合素的表达和相互作用在肾小球系膜区,肾间质及GBM的形成。 Fibronectin (FN) and its integrin receptor play an important role in embryogenesis by regulating cell migration, proliferation and differentiation. In this study, we examined, by immunohistochemistry, the temporal and spacial expression of FN and β_1 integrin receptor (α5 subunit) in fetal and adult kidney. Our data showed that FN and α5 chain of integrin only weakly expressed around the ureteric bud cells in the early stage of developing kidney. As the kidney maturing, the glomerular mesangium was immunoreactive, and was intensely positive after the nephron undergoing capillary loop expansion. In addition, the antibodies to FN and α_5 chain was strongly labeling in GBM, predominantely the segments nearby the vascular pole in adult kidney, as well as in mesangium and interstitial. The capillary wall in interstitial was also markedly immunoreactive to them. These findings suggest that the expression and interaction of FN and α_5β_1 integrin regulate the formation of mesangium, GBM and interstitial, and play a key role in the maturation of human kidney.
