红花注射液对阿霉素所致大鼠心肌损伤保护作用 被引量:5

摘要 目的观察红花注射液对阿霉素诱导大鼠心肌损伤的保护作用和抗氧化作用。方法选用阿霉素诱导大鼠心肌损伤模型。SD大鼠60只,随机分为3个组,每组20只,分别为对照组、损伤组、治疗组。对照组:实验第1~9天注射生理盐水,每天3mL/kg,l次/d。治疗组:实验第1~9天注射红花注射液,每天2g/kg,1次/d,第4天注射阿霉素,隔天1次,连用3次,用生理盐水配置成lmg/mL,每次3mg/kg。损伤组:实验1~9天注射生理盐水,每天3mL/kg,1次/d。第4天注射阿霉素,以后隔天1次、连用3次,用生理盐水配置成1mg/mL,每次3mg/kg。实验结束测定血清CK,SOD活性及心肌匀浆MDA含量、AST活性,并进行心肌病理检查。结果经阿霉素损伤后,血清CK活性及心肌匀浆MDA含量增高,AST活性增高,SOD活性降低,与正常组比较有极显著性差异(P<0.01);心肌MDA含量降低,CK及AST活性降低,SOD活性升高,与损伤组比较均有显著性差异(P<0.01)。结论红花注射液有抗氧化作用和对阿霉素所引起的心脏毒性具有保护作用,为临床寻找有效的阿霉素所致心肌损伤保护药物提供良好的客观依据。 Objective To observe the protective effect and antioxidative effect of Honghua injection (HHI) on the myocardiam injury induced by adriamycin(ADM) in rats. Methods Sixty SD rats(weighing 200-220g) were randomly divided into three groups. In the control group,normal saline was ip injected at a dose of 3mL/kg once a day for 9 days. The injury group was given ip injection of ADM at a dose of 3mg/kg from the 4th day,once every other day for 3 times and normal saline at a dose of 3 mL/kg once a day for 9 days. The therapy group was given ip injection of ADM at a dose of 3mg/kg from the 4th day,once every other day for 3 times and HHI at a dose of 2g/kg once a day for 9 days.The activity of superoxided lsmutase(SOD), serum creatine phosphokinase activity(CK) and the level of malondialdehyde (MDA) were assayed,and at the same time,the rat myocardial pathomorphism was detected. Results Compared with the injury group,the serum SOD activity was higher(P〈0.01),and the serum MDA activity was lower in therapy group (P〈0.01). The protection of myocardial pathological damage in therapy group was improved (P〈0.01). Conclusion The HHI has protective effect and antioxidative effect on the myocardium injury induced by adriamycin in rats.
