中药肝心宁对CCl_4诱导的肝纤维化大鼠TGF-β1/Smad信号通路的影响 被引量:4

Effects of Traditional Chinese Medicine Ganxinning on TGF-β1/Smad Signaling Pathway in Carbon Tetrachloride-induced Hepatic Fibrosis of Rats

摘要 目的:研究中药肝心宁对CCl4诱导的大鼠肝纤维化肝脏TGF-β1及Smad-3表达的影响,探讨其抗纤维化的作用机制。方法:采用四氯化碳皮下注射并饲以高脂低蛋白饲料和30%酒精诱导大鼠肝纤维化模型,68只雄性SD大鼠随机分成5组:正常组(11只)、模型组(15只)、阳性对照组(14只)、肝心宁高剂量组(14只)和肝心宁低剂量组(14只),6周末处死所有动物,行HE染色,光镜下观察肝组织肝纤维化程度,并采用RT-PCR半定量方法,检测大鼠肝组织TGF-β1 mRNA、Smad-3mRNA的表达水平。结果:与正常对照组相比,模型组TGF-β1 mRNA、Smad-3 mRNA表达明显增强。经肝心宁治疗后大鼠肝脏TGF-β1表达比模型组减弱,肝心宁同时下调Smad3的表达。另外,肝心宁组大鼠肝脏病理变化显著改善。结论:肝心宁可下调TGF-β1 mRNA、Smad-3 mRNA蛋白的表达,从而抑制或减轻肝纤维化的形成。 Objective: To study the effects of Ganxinning on TGF - β1 and Smad3 expressions of the experimental hepatic fibrosis induced by carbon tetrachloride (CC14) in rats, and to explore its anti -fibrotie molecular mechanism. Methods: Sixty -Eight male SD rats were rando mLy divided into 5 groups: normal group (N group, 11 rats), model group M group, 15 rats), positive control group (PC group, 14 rats) , high dose of Ganxinning group (GXNh group, 14 rats) and low dose of Ganxinning group (GXN1 group, 14 rats) . The rats of M, PC, GXNh and GXN1 group were disposed with mixture of CC14 and peanut for inducing liver fibrosis, sc twice a week , at the same time which were feeded with high - fat and low - albumen food and 30% alcohol. The GXNh or GX- N1 group was also treated with high dose or low dose of Ganxinning , respectively 2.6g/100g · mL/d or 0. 65g/100g? mL/d , ig once a day ; the PC group was treated with colchicines , ig once a day ; the M group were given singly - distilled water , ig once a day ; the N group were not disposed. At the end of 6 weeks all the survived rats were sacrificed. The pathological changes of liver were ob- served by H- E staining , TGF- β1 mRNA and Smad -3 mRNA were detected by RT- PCR. Results: In comparison with model group, the levels of TGF - β1mRNA were significantly decreased after treatment with Ganxinning. Meanwhile, the levels of Smad -3 mRNA was also down - regulated. The pathological changes of liver tissues in Ganxinning groups were also markedly alleviated. Conclusion: Ganxinning significantly inhibit the development of liver fibrosis in rats induced by CC14 , and its underlying mechanism might relate to decreasing the expression of TGF - β1 and Smad - 3.
