摘要
为提高酵母RNA提取效果,采用超声辅助提取工艺,在单因素实验的基础上,采用超声处理时间、超声功率和提取时间进行三因素三水平响应面分析实验以优化此工艺条件。结果表明,酵母RNA超声辅助提取的最佳工艺条件为超声处理时间10min,超声功率300W,提取时间20min,在此工艺条件下,酵母RNA实际得率为9·56%,与模型预测值之间具有较好的拟合性。在3个因素中,超声处理时间、超声功率对酵母RNA得率的影响极显著,提取时间影响不显著,且相互之间无交互效应。
In order to optimize ultrasound-assisted extraction technology for the extraction of RNA from yeast, on the basis of single factor experiments ,the effects of operating conditions, such as ultrasonic time, ultrasonic power and extraction time on the yield of RNA from yeast were analyzed by response surface methodology.The optimized conditions of ultrasound-assisted extraction were as follows;ultrasonic treatment time was lOmin,ultrasonic power was 300W, and extraction time was 20min.Under the above-mentioned conditions,the actual yield of yeast RNA was 9.56%,which is well matched with the predicted yield.Among three factors, ultrasonic treatment time and ultrasonic power had significant effects on the yield of yeast RNA, while extraction time had no remarkable effect.Moreover,there are no interaction among three factors.
