摘要
研究了相思愈伤组织继代培养过程中,激素浓度、光照条件、接种量等各因素对相思愈伤组织培养的影响。结果发现,在培养基中添加0.2~0.5mg/L的2,4-D浓度水平最有利于相思愈伤组织的生长。通过不同光照强度的对比试验得出,弱光在保持愈伤组织的形态上较有利。将接种量控制在0.3g/瓶时,在愈伤组织的增殖和保持能达到最优的效果。同时,通过对不同相思愈伤组织细胞学观察可以看出,不同相思品种的愈伤组织细胞形态各不相同,在培养过程中,愈伤组织的细胞的形态也在变化。
The different effects such as plant growth regulators, light conditions, and genotypes on the maintenance of the three species of Acacia were compared in the experiment, and the results showed that the medium supplemented with 0.2-1 mg/L 2,4-D was beneficial to the maintenance of callus. Moreover, the callus could be kept loose and soft under the weak light by comparing the effects of different light conditions. And the superior inoculation amount was 0.3 g callus in each bottle. By the histological observation on different callus in acacia, it showed that there were different Morphological cells and callus in different species of Acacia which also changed during the culture.
