摘要
目的:为了探讨中华眼镜蛇毒(NNAV)对荷人鼻咽癌(NPC)裸鼠血浆丙二醛(MDA)和超氧化物歧化酶(SOD)活性的影响。方法:采用比色法检测经腹腔注射低、中或高浓度NNAV(1μg/ml、5μg/ml、10μg/ml)的荷人NPC裸鼠血浆MDA和SOD活性。结果:荷瘤鼠血浆MDA(842nmol/ml)比正常裸鼠(392nmol/ml)明显升高,而荷瘤鼠血浆总SOD(T-SOD)、锰SOD(Mn-SOD)和铜锌SOD(CuZn-SOD)活性(5219Nu/ml,2031Nu/ml,3188Nu/ml)比正常裸鼠(12564Nu/ml,75.11Nu/ml,50.53Nu/ml)低;低、中和高浓度NNAV均可使荷瘤鼠血浆MDA降低或至正常范围(562nmol/ml,5.17nmol/ml,4.58nmol/ml),并可使荷瘤鼠T-SOD、Mn-SOD和CuZn-SOD有不同程度的升高,其中以高浓度组为佳(10888Nu/ml,7227Nu/ml,3661Nu/ml,)。结论:结果提示NNAV的抑瘤作用可能与其降低荷瘤鼠自由基水平和提高SOD活性有关。
Objective:To investigate the changes of plasma malondialdehyde(MDA)and superoxide dismutase(SOD)activitg in nude mice(NM)bearing human nasopharyngeal carcinoma(NPC)and observe the effects of Naja Naja atra(NNAV) on them.Methods:Plasma MDA and SOD in human NPC-bearing NM treated by low、 middle or hight-concentration NNAV solution (l μg/ml,5 μg/ml,10 μg/ml)peritoneally were determined by colorimetry.Results:Plasma MDA content(8.42 nmol/ml) in NM bearing tumor increased markedly in comparison with the control group (3.92 nmol/ml).Whereas plasma T-SOD、Mn-SOD and Cu-Zn-SOD activity (52.19 Nu/ml,20.31Nu/ml,31.88 Nu/ml) in NM bearing tumor control group(125.64 Nu/ml,75.11 Nu/ml,50.53 Nu/ml) decrease significantly in comprison with blank control group. Low,middle or high-concentration NNAV solution(especial hight-concentration group) decreased plasma MDA content and enhanced SOD activity in NM bearing tumor. Conclusion:The antitumor effect of NNAV might be by means of decreasing free redicals and increasing SOD activity in NM bearing tumor.
