摘要
目的观察葛根素对糖尿病大鼠视网膜细胞间黏分子-1(ICAM-1)表达的影响。方法利用链脲佐菌素腹腔注射法建立糖尿病大鼠模型,将实验用Wistar大鼠随机分为正常对照组、葛根素治疗组和糖尿病组,治疗12周。观察治疗期间及治疗后各组大鼠的一般情况、尿糖、血糖、血NO及血SOD值,HE染色及免疫组化染色法观察大鼠视网膜形态及ICAM-1表达的不同。结果和正常对照组相比,造模组大鼠血糖均升高,血SOD和血NO值降低(P<0.05)。葛根素能改善大鼠基本情况,降低血糖(P<0.05),升高血SOD及血NO值(P<0.05),并减少视网膜组织细胞间ICAM-1表达,其差异有统计学意义。结论葛根素对糖尿病大鼠视网膜组织间ICAM-1的表达具有抑制作用,从而对高血糖引起的视网膜损伤有一定的保护作用。
Objective To observe the Effect of puerarin on ICAM -1 in streptozotocin-induced diabetic rats'retina. Methods Diabetic rats were induced by intraperitoneal injection of STZ. Rats were randomly allocated into normal control group, puerarin group , and diabetic group. The puerarin treatment lasted for 12 weeks. During and after the treatment,the general health conditions, blood glucose levels, urine glucose levels, NO value of blood, SOD value of blood were obseived. Hematoxylin-Eosin staining and Immunohistochemistry were used to observe the rat's retinal and detect the different expression of ICAM- 1 protein in the retina. Results Compared with the normal control group, blood glucose levels increased and NO value of blood , SOD value of blood decreased in the puerarin group and diabetic group(P〈0.05). Puerarin could improve the general health of the rats, decreasing blood glucose levels (P〈0.05), increasing NO value of blood and SOD value of blood (P〈0.05), reducing the expression of ICAM- 1 in the retina. The difference is significant. Conclusion Puerarin may inhibit the expression of ICAM- 1 in the diabetic rats'retina, and protect the retina by reducing the blood glucose levels.
