急性视网膜坏死患者玻璃体MMP-2和MMP-9活性研究

摘要 目的检测急性视网膜坏死(acute retinal necrosis,ARN)患者玻璃体内基质金属蛋白酶(matrix metalloproteinases,MMPs)2和9的活性。方法6例ARN患者行玻璃体切割术抽取玻璃体液,6例单纯孔源性视网膜脱离(rhegmatogenous detachment,RD)患者作为对照,采用明胶酶谱法进行MMP-2和MMP-9活性检测。结果RD患者玻璃体标本显示72k DaMMP-2酶原和62k DaMMP-2活性酶条带,无明显MMP-9条带成分,ARN患者标本可见到92kDa MMP-9酶原、82kDaMMP-9活性酶、72k DaMMP-2酶原和62k DaMMP-2活性酶条带,ARN组所有条带的表达都要显著高于RD组(P<0.01)。结论MMP-2和MMP-9可能在ARN的玻璃体视网膜炎症反应中起到非常重要的作用。 Objective To detect the activity of matrix metalloproteinases (MMP) 2 and 9 in vitreous of patients with acute retinal necrosis (ARN). Methods Vitreous samples from 6 cases with ARN were obtained in process of vitrectomy. Samples from 6 cases of rhegmatogenous detachment of retina (RD) without PVR were set as control. Activities of MMP-2 and -9 were detected by gelatin zymography. Resuits Only 72kDa zemogen and 62 active enzyme of MM-2 could be detected in control, while 92kDa zemogen and 82 active enzyme of MM- 9 and 72kDa zemogen and 62 active enzyme of MM-2 could be detected in vitreous of ARN. Concentrations of all four bands were significantly higher in ARN patients than that in control (P〈0.01). Conclusion Gelatinase A (MMP-2) and B (MMP-9) might play important roles in inflammatory responses of ARN patients.
