摘要
目的观察体外乙醇对斑马鱼血管内皮细胞生长因子受体2(VEGFR2)基因表达的影响。方法选取绿色荧光蛋白特异性标记血管内皮细胞的转基因斑马鱼胚胎120颗,随机均分为1%乙醇处理组(A组)、1.5%乙醇处理组(B组)、1.75%乙醇处理组(C组)及对照组(D组)。于胚胎受精后4.3h~3d给予一定体积浓度比的无水乙醇处理,制备胎儿酒精综合征模型。用激光共聚焦显微镜观察其荧光表达的情况,并用反转录PCR方法观察乙醇处理后VEGFR2的表达。结果与D组比较,A、B、C组斑马鱼胚胎在受精后3d,荧光强度和VEGFR2表达随乙醇浓度增高而减弱,表达下降。结论斑马鱼胚胎受精后3d,VEGFR2基因表达随着乙醇浓度的增加而减弱。
Objective To investigate the expression of vascular endothelial growth factor receptor 2(VEGFR2) gene in zebrafish embryo development induced by in vetro ethanol with different concentration.Methods One hundred and twenty embryos were randomly divided into four groups with 30 embryos each,which were treated with different ethanol 1% (group A), 1.5% (group B), 1.75%(group C), concentrations(group D) (v/v) between 4.3 hours and 3 days after fertilization. Confocol microscopy and reverse transcriptase-PCR were used to observe the fluorescent intensity and mRNA expression of VEGFR2. Results Compared with group D, the fluorescent intensity and VEGFR2 expression on the 3^nd day after fertilization decreased as the ethanol concentration increased. Conclusion Treated with in vetro ethanol, the expression of VEGFR2 was decreased dose- dependently.
