摘要
目的观察exendin-4(Ex)对链脲佐菌素(STZ)诱导的糖尿病小鼠模型胰腺survivin基因表达的影响,旨在探讨ex-endin-4抗凋亡作用的机制。方法6 wk ♂ BALB/c小鼠分为3组:STZ组、Ex组(STZ+Ex)、对照组。每周测量血糖,4wk时胰腺行免疫组化、TUNEL检测,实时荧光RT-PCR方法检测胰腺survivin表达水平。结果Ex组血糖低于STZ组,胰岛中胰岛素染色阳性面积百分比大于STZ组(39.40±9.72vs25.47±9.26,P<0.01),胰岛凋亡细胞数目少于STZ组。STZ注射后第4周胰腺survivin mRNA水平升高,Ex组更高,差异有统计学意义(P<0.05)。结论Ex可抑制STZ诱导糖尿病小鼠的胰岛细胞凋亡,此作用可能部分通过上调胰腺survivin表达介导。
Aim To investigate the effect of exendin-4 (Ex) on the expression of survivin in pancreas in streptozotocin (STZ)-induced diabetic mice. Methods Balb/c mice (6-weak-old) were divided into three groups : STZ group, Ex group and control group. Blood glucose concentrations, immunocytochemistry and trans- ferase-mediated dUTP nick-end labeling assay (TUNEL) were performed in the present study. Expression of survivin mRNA was detected by real time PCR. Results Blood glucose concentration in Ex group was lower than that in STZ group during the experimental period. At the end of experiment, the percentage of insulin positive area in Ex mice were higher than that of STZ mice ( 39. 40 ±9.72 vs 25.47 ± 9.26, P 〈0. 01). Apoptotic islet cells in Ex group were decreased than those in STZ group. Expression of survivin gene in pancreas was significantly increased at the end of 4th week after STZ administration, and it was higher in Ex group than in STZ group. Conclusion Exendin-4 reduces islet cell apoptosis in STZ-induced diabetic mice, which is partially mediated by up-regulating expression of survivin.
