紫外线诱导家蚕胚胎细胞BmE-SWU1的凋亡研究 被引量:3

Study on Apoptosis of Silkworm Embryo Cell Line BmE-SWU1 Induced by Ultraviolet Irradiation

摘要 UVB诱导家蚕胚胎细胞BmE-SWU1不同时间后,用四氮唑盐法(MTT)和Hoechst染色及DNA-Ladder法分别检查BmE-SWU1细胞的增殖活性和凋亡情况.该实验研究了紫外线诱导对家蚕胚胎细胞BmE-SWU1凋亡的影响,建立了紫外线诱导家蚕细胞凋亡的简单模型,为家蚕变态发育过程中细胞凋亡的研究奠定了基础.结果表明:UVB处理家蚕BmE-SWU1细胞后,细胞增殖活性随着照射剂量的增加而逐渐降低;光学显微镜下可见细胞膜表面突出或出现小泡,经Hoechst染色后,凋亡小体在荧光显微镜下清晰可见,细胞DNA电泳条带呈梯形分布,这是凋亡细胞典型的生化特征. In order to study the influence of UVB (ultraviolet-B) on apoptosis of silkworm cell BmE SWU1, the BmE-SWU1 cells were induced with UVB and their proliferation and apoptosis were examined with MTT, Hoechst staining and DNA-Ladder. After irradiation of BmE-SWU1 cells with UVB, the proliferation activity of the cells dropped with increasing irradiation doses. Light microscopic examination re vealed tiny bubbles on the surface of the cell membranes. After staining by Hoechst, distinct apoptotic bodies could be seen under the fluorescence microscope and the DNA fragments of BmE-SWU1 cells showed a ladder-shaped distribution in agarose gel electrophoresis-a typical characteristic of cell apoptosis. It is thus concluded that UVB radiation can induce apoptosis in BmE-SWU1 cells.
