摘要
为探索蛋白质溶液中的热原物质去除问题 ,采用氢氧化铝吸附法 ,选择较常见的牛血清白蛋白 (BSA)、牛血红蛋白 (BH)和溶菌酶 (LYZ)为模型 ,较详细研究不同蛋白质溶液中氢氧化铝的吸附效率及蛋白回收问题。结果表明 ,氢氧化铝配比、蛋白质种类、蛋白质浓度、内毒素(ET)含量、酸碱度、离子强度等因素都显著影响吸附效率。
In order to resolve the problem that how to remove the pyrogen in protein solution, three kinds of proteins bovine serum albumin (BSA), bovin hematoglobin (BH) and lysozyme (LYZ) were chosen as a model, the Aluminium Hydroxide was used to adsorb the endotoxin (ET) in above protein solution. The adsorption rate and protein recovery were studied in different protein solution. The results showed that the ratio of Aluminium Hydroxide, the type of protein, the protein concentration, the endotoxin level, pH, the ionic strength, etc, all of them effected the adsorption apparently. These results could provide scientific basis for the best removing pyrogen technology in the course of pharmacy protein processing.
