稀有放线菌噬菌体ΦSCU20的分离及其生物学特性的研究

摘要 以稀有放线菌小单孢菌40027菌株为指示菌,从土壤中分离得到1株新噬菌体,命名为ΦSCU20。在测试的9株放线菌菌株中,仅能感染小单孢菌40027菌株和A-M-01菌株。电镜观察显示噬菌体ΦSCU20由多面体的头部和尾部组成。噬菌体ΦSCU20生物学特性研究表明,噬菌体ΦSCU20形成噬菌斑时培养基中适宜的Ca2+和Mg2+浓度分别为8 mmol/L和20 mmol/L;噬菌体ΦSCU20在储存液中适宜的pH值为8;噬菌体ΦSCU20不耐高温,经40℃保温30 min后,其活力降低到48.65%,经60℃保温30 min后,则完全失活;噬菌体ΦSCU20基因组特性研究表明,该噬菌体的基因组为双链DNA,基因组大小约为65 kb。 Rare actinomycete phage ΦSCU20 was isolated from soil using Micromonospora sp. 40027 as indicator strain. Phage ΦSCU20 could infect both Micromonospora sp. 40027 and Micromonospora A-M-01. Phage ΦSCU20 has the hexagonal heads and tails; the plaque formation of phage ΦSCU20 on Micromonospora sp. 40027 was the best on DNA medium with addition of 8 mmol/L Ca^2+ and 20 mmol/L Mg^2+ ; Phage ΦSCU20 was stable at pH 8; the suitable incubation temperature for the propagation of ΦSCU20 was 28 ℃; 48. 65% of ΦSCU20 remained viable when it was incubated at 40 ℃ for 30 rain. Restriction digestion analysis of genomes of phage ΦSCU20 indicated that it was double-stranded DNA and its genome size was 65 kb.
