Ⅳ型胶原粘附法纯化富含干细胞的鼠切牙颈环上皮细胞 被引量:2

Isolation of tooth epithelial stem cells in rat lower incisors by the adhesive capacity to type Ⅳ collagen

摘要 目的:探讨Ⅳ型胶原对实验室培养条件下富含干细胞的鼠切牙颈环上皮细胞纯化的影响。方法:以Ⅳ型胶原包被培养瓶,再接种颈环上皮混杂细胞,以未包被胶原的培养瓶做对照,观察贴壁细胞的生长形态;用β1整合素和CK角蛋白对贴壁细胞进行免疫组织化学染色鉴定。结果:与对照组相比,Ⅳ型胶原包被组纤维样细胞被完全抑制,贴壁生长的细胞全部为上皮细胞,形成大的克隆,β1整合素和CK角蛋白呈阳性表达。结论:Ⅳ型胶原粘附法可以获取较纯的鼠切牙颈环上皮细胞。 Objective:To explore the effect of type Ⅳ collagen on purification of cervical-loop epithelial cellular of Wistar rat lower incisors. Method:Flasks coated type Ⅳ collagen as experimental group with coated nothing as control group. The primary cuhures,a mixture of different cells,were seeded in the flasks respectively. The cells adhering to collagen type Ⅳ were identified by immunohist-chemistry staining. Result:Compared with the control group,fibre-like cells were fully removed in the experimental group.The cervicalloop epithelial cells were poly-angle and clone formation.The cervical-loop epithelial cells exhibit positive expression for integrin-β1 and monoclonal antibody CK in immunohistochemistry staining. Conclusion:The purified cervical-loop epithelial cells in rat lower incisors could be obtained by adhering to collagen type Ⅳ.
