豆粕水解液为原料根霉产纤溶酶液体发酵培养基的优化 被引量:2

Optimization of liquid fermentation media of fibrinolytic enzyme produced by Rhizopus chinesis using soybean cake hydrolysate as substrate

摘要 以豆粕水解液为原料,研究根霉产纤溶酶液体发酵培养基的组成。实验结果表明,豆粕的最适水解时间为1.5 h,麸皮水与豆粕水解液的最适体积比例为2∶3,金属离子对酶活力提高无显著作用,最适培养基配方是:5%麸皮水,6%豆粕水解液,0.1%氯化铵,0.2%硝酸钠,2.5%胰蛋白胨,初始pH4.3,酶活力为177.62 U/mL。 The components of liquid fermentation medium of fibrinolytic enzyme produced by Rhizopus chinesis were studied,in which soybean cake hydrolysate was used as substrate. The optimal conditions were obtained as followed : hydrolysis time of soybean cake 1.5 h, the ratio of wheat bran water and soybean cake hydrolysate 2:3. The metal ions had no significant effects on enzyme activity. The optimal medium composition was as followed: wheat bran water 5% ,soybean cake hydrolysate 6% ,NH4Cl 0. 1% ,NaNO3 0. 2% ,tryptone 2. 5% ,initial pH 4. 3. Under these conditions, the activity reached 177. 62 U/ml.
