摘要
目的:研究严重烧伤和热休克处理大鼠肾脏热休克蛋白72(HSP72)基因表达,并讨论其作用和意义。方法:采用HSP72特异性引物,利用反转录聚合酶链式反应(RT-PCR)和原位杂交技术观察肾脏HSP72mRNA变化;用免疫印迹观察蛋白质表达的变化。结果:①烧伤早期大鼠肾脏热休克蛋白表达是逐渐增加的,到24h,其mRNA和蛋白质均处于高表达水平。②热休克预处理后大鼠肾脏热休克蛋白表达峰值在6h~12h,在24h其表达水平仍比正常对照高。③HSP72mRNA在近曲小管、远曲小管、集合管和髓拌上皮细胞均有分布。结论:烧伤后肾脏hsp72mRNA表达较热预处理后时间长,HSP72分布广泛。
Objective: The expression of heat shock protein 72 gene (HSP 72) in the renal tissue was observed after severe burn and heat shock in rats and its effects and significance discussed. Methods: The expression of HSP 72 mRNA was determined with RT-PCR, the expression of HSP 72 protein with western blotting and its distribution with hybridization in situ. Results: ①After burn injury, the expression of HSP 72 mRNA and protein was increased progressively in the early stage and it reached a high level in the 24th hour postburn and remained at that level since then.②The accumulation of HSP 72 mRNA and protein reached the peak in the period from 6th to 12th hour in the kidneys. Even in the 24th hollr, the level of HSP 72 mRNA and protein remained quite high. ③In the rat kidneys, HSP 72 mRNA distributed in the epithelial cells of the proximal and distal convoluted tubules and collecting tubes. ConclUSion: The expression of HSP 72 gene after burns remained longer than heat shock and its distribution was quite wide with high expression level.
