摘要
将银中杨(Populus alba×P.berolinensis)无菌苗的叶片接种到MS+0.2mg/L TDZ+0.02mg/LNAA+30g/L蔗糖中培养,于15d左右诱导出不定芽,30d左右形成小苗。对叶片不定芽再生过程的生理生化变化的测定结果表明:在不定芽形成过程中,在0~15d的培养周期内,POD、SOD活性、可溶性蛋白及可溶性糖含量在开始阶段均呈上升趋势,在叶片不定芽形成的高峰期15d时,各项指标均达到最高值,随后呈不同程度地下降。
The leaf explants from in vitro cultured plantlets of Populus alba × P. berolinensis were cultured on MS medium supplemented with 0.2 mg/L TDZ, 0.02 mg/L NAA, and 30 g/L sucrose. The explants began to produce adventitious shoots after 15 days of culture, and the regenerated plantlets occurred after 30 days of culture. The contents of soluble protein, soluble sugar, and the activities of POD and SOD first increased and then decreased during the adventitious shoot formation with 15 days of culture. Results demonstrate that adventitious shoot regeneration is strongly associated with physiological changes.
